简要概述

锌离子荧光探针Zinquin 乙酯是美国AAT Bioquest生产的用于检测锌离子的荧光探针,Zinquin乙酯是亲脂性的,锌敏感的,可透过细胞的荧光探针。它保留在活细胞中,因为乙酯被细胞溶质酯酶切割,产生携带负电荷的Zinquin,阻止其流过质膜。Zinquin荧光探针可以通过在含有5-40μMZinquin乙酯的PBS中与钙和镁(或在培养基中)的培养基中加载到细胞中。通常将细胞与Zinquin乙酯在37℃下孵育15-30分钟。确切的加载浓度,时间和温度取决于实验目的和细胞类型,因此需要通过实验优化。用PBS在培养基中洗涤细胞以除去细胞外剩余的染料。在显微镜下观察细胞或用于共聚焦显微镜,FACS或荧光分光光度法分析。锌是体内第二丰富的过渡金属,它作为催化,结构和调节离子必不可少。锌离子参与体内平衡,免疫反应,氧化应激,细胞凋亡和衰老。已经提出锌作为突触前神经元的常规神经递质和跨越突触后神经元的跨膜信号起作用。异常的锌代谢与许多神经系统疾病有关,包括阿尔茨海默病,帕金森病和癫痫。已证明最适合体内监测锌的技术是荧光成像。金畔生物是AAT Bioquest的中国代理商,为您提供最优质的锌离子荧光探针Zinquin 乙酯。

产品说明书

操作步骤

 1.在DMSO中制备~10mM储备溶液。
 2.装入含有5-40μMZinquin乙酯或PBS(含钙和镁)的培养基中的细胞中。
 3.在37ºC下与Zinquin乙酯一起孵育15-30分钟。确切的加载浓度,时间和温度取决于实验目的和细胞类型,因此需要通过实验优化。
 4.用PBS洗涤以除去细胞外剩余染料,并用PBS或培养基替换。
 5.在显微镜下观察或用流式细胞仪分析。
