荧光素醛是一种活性荧光染料,可与胺、肼或羟胺反应。 荧光素是应用最广泛的绿色荧光染料。 醛基对 pH 5-9 的胺、酰肼或羟胺基具有反应性。 与胺反应性琥珀酰亚胺酯基团 (NHS) 不同,醛可以在酸性 pH 下与 N 端胺基反应,某些生物偶联反应有时需要这种条件。 醛与胺基反应形成中间席夫键。 用氢化物进一步还原将形成稳定的 C-N 键。 醛和其他基团之间的反应允许荧光素染料在目标分子上的所需位置进行位点特异性结合和标记。 该染料可以通过 FITC 通道下的普通荧光仪器轻松检测。
