产品介绍

TWO-PRO -1在化学上等同于TO-PRO®-1(TO-PRO®是Invitrogen的商标)。TWO-PRO -1是一种碳花青二聚体,具有类似于FITC的绿色荧光。它不具有细胞渗透性,可以很容易地与Cy5和罗丹明区别开来,作为核复染和死细胞指示剂。在不存在核酸的情况下它是非荧光的,并且在与DNA结合后会产生非常明亮的荧光信号。TWO-PRO -1可在培养细胞和石蜡切片中进行强而有选择性的核染色。同时使用细胞可渗透的Nuclear Red LCS1染料和不渗透细胞的TWO-PRO -1进行标记,可用于评估细胞活力。TWO-PRO -1和Nuclear Red 的消光系数都比结合DNA的碘化丙啶大。

简要概述

蛋白酶荧光底物Gly-Pro-AMC是美国AAT Bioquest生产的荧光染料,Gly-Pro-AMC是用于检测二肽酰多肽酶IV (DPPIV)的高灵敏度荧光底物,一种丝氨酸蛋白酶,能够从倒数第二位上是L-脯氨酸或者L-丙氨酸的多肽上剪切其N末端的二肽酰。DPPIV是表达于多种细胞(包括上皮细胞,内皮细胞和淋巴细胞)表面的多功能蛋白质。它与T细胞活化抗原CD26和腺嘌呤核苷脱氨基酶(腺苷脱氨酶)结合蛋白具有同一性,它以可溶性状态释放于血浆中。CD26/DPPIV的底物包括多种含有脯氨酸的多肽,例如生长因子、趋化因子、神经肽和血管活性肽。DPPIV参与免疫调控,信号转导和细胞凋亡,并且在肿瘤的发展中发挥重要作用。重要的是,DPPIV是II型糖尿病的治疗靶点,由于其作为血清蛋白酶,能够剪切多肽胰高血糖素家族中肠降血糖素荷尔蒙,以此调节体内葡糖糖的稳定。研究显示一种DPPIV抑制剂可以增强葡糖糖衰减的耐受力。用带有Gly-Pro-AMC的DPPIV检测是一种简单、快速,灵活的检测法,是理想的高通量筛选方法。与DPPIV检测法相比,这种检测法增强了检测灵敏度,使研究者可以用更少的酶,同时获得恰当的Z值。这种单一性的酶偶联形式使用非常方便,只需要在检测样品中添加一种单一的试剂;在20inm内可以获得最大感光值,并且其单一产物非常稳定。金畔生物是AAT Bioquest的中国代理商,为您提供最优质的蛋白酶荧光底物Gly-Pro-AMC。

产品说明书

样品实验方案 

溶液制备 

1.储备溶液

所有未使用的储备溶液应分为一次性使用的等分试样,并在制备后储存在-20°C下。 避免重复冻融循环。
1.1 Gly-Pro-AMC:
在DMSO中制成10至25 mM的Gly-Pro-AMC储备溶液,避光。 注意:储备溶液应及时使用; 应将所有未使用的溶液等分,并在-20℃冷冻。注意:避免重复冻融循环,并避免光照。

 

2.工作溶液

Gly-Pro-AMC:
取出一个小瓶,并用您选择的测定缓冲液(例如50 mM Tris-HCl,pH 7.5,1.0 mM DTT)稀释储备溶液,制成50至100 µM测定溶液。

 

样品操作及分析

1.将等体积的DPPIV标准品或样品与测定溶液混合,并在室温下孵育至少1小时。

2.在Ex / Em = 380/500 nm处监测荧光增加。
