样品分析方案

概述

细胞或组织培养

在封闭缓冲液中添加一抗

加入结合HRP的二抗

准备Styramide™(PSA)或Tyramide(TSA)工作溶液,并在室温下在细胞或组织中涂抹5-10分钟 

 

储备溶液配制

1.PSA储备溶液(100X)

将100μLDMSO加入到iFluor™染料标记的Styramide™缀合物(例如#45000)的小瓶中,制成100X Styramide™储备液。
注意:制成单次使用的等分试样,并将未使用的100X储备溶液在2-8°C的暗处存储,并避免重复冻融循环。

2. H2O2储备溶液

向90μL的ddH 2 O中添加10μL的3%过氧化氢(组分B)。
注意:在使用当天准备新鲜的100X H 2 O 2溶液。

 

工作溶液配制

PSA工作溶液(1X)

每1 mL PSA工作溶液需要1 mL TSA / PSA缓冲液(组分A),10μLStyramide™储备溶液和10μLH 2 O 2储备溶液。
注意:1 mL足够进行10次测试,基于每个盖玻片或96孔微孔板中每孔所需的100μLStyramide™工作溶液。
注意:Styramide™工作溶液必须在制备后2小时内使用,并避免直接暴露在光线下。
