荧光抗体缀合物在紧密接触时趋于聚集,由于FRET和不同缀合物之间的其他能量转移现象,扭曲了多色染色细胞的分析结果,使荧光染料相互作用会导致染色伪影,从而让数据复杂化。 CytoWatch 多路复用缓冲液是为细胞的多色流式细胞分析而研发的缓冲液。 CytoWatch多路复用缓冲液是一种浓缩溶液,可在染色细胞之前直接添加到某些荧光抗体缀合物或其他荧光试剂的染色溶液中。 它可用于增强或改进利用两种或更多种不同染色试剂(例如,与BV染料或Qdot缀合的抗体)的多色流式细胞术实验。

产品介绍

海藻糖是一种天然存在的二糖,在某些单细胞和多细胞生物物种中具有超强的抗逆能力。在哺乳动物细胞中,细胞内和细胞外海藻糖的存在已被证明可以提高对冷冻和干燥的耐受性。由于哺乳动物细胞既不合成也不导入海藻糖,因此有效地细胞内递送海藻糖的新方法的开发一直在进行中。在这里,我们研究了工程海藻糖亲脂性衍生物的膜渗透性。与常规的海藻糖,海藻糖二乙酸酯(2-O-Ac-Tre)和海藻糖四乙酸酯(4-O-Ac- Tre)。一旦进入牢房 细胞内酯酶水解6-O-Ac-Tre分子,将游离海藻糖释放到细胞质中。细胞内海藻糖(加上乙酰化变体)的总浓度高达6-O-Ac-Tre细胞外浓度的10倍,达到适用于生物保存的浓度。为了描述这种累积现象,提出了扩散反应模型,并通过拟合实验数据确定了6-O-Ac-Tre的渗透性和反应动力学。进一步的研究表明,负载量和细胞内海藻糖的存在对细胞活力和功能的影响可以忽略不计。设计海藻糖化学结构而不是操纵细胞是一种无害的,细胞友好的海藻糖递送方法,
